RESUMEN

The manufacture of wind turbine blades generally uses balsa wood as the base materials, and it is crucial to explore new regions for cultivating balsa trees to achieve carbon neutrality in the future. Xishuangbanna may be China's only area with a tropical climate suitable for the large-scale planting of balsa trees. The present study investigated the key soil elements influencing the growth of balsa plantations and the effects of different cultivation practices on soil environments and economic benefits in Xishuangbanna, China. We found that the height of balsa stems after growing 4 years reached 5.8 m; the increment of diameter at breast height (DBH) reached 27.7 cm and volume of balsa stems reached 196.0 m3 ha-1 in Xishuangbanna of China. It is of the utmost importance to improve the contents of soil exchangeable magnesium (Mg) and available phosphorus (P) for the growth of balsa trees, and exchangeable aluminium (Al) inhibited the growth of balsa trees. The practice of plastic film mulching not only improved soil moisture in the 40‒100-cm soil layer in the dry season and in the 0-60-cm soil layer in the rainy season but also enhanced soil nitrate nitrogen when compared with no plastic-mulching practice in balsa plantations. The comprehensive economic benefits of balsa/coriander/ginger/taro plantations were significantly improved by implementing plastic film mulching, as compared to balsa plantations. We conclude that balsa tree can be cultivated in Xishuangbanna, China, and its successful cultivation provides opportunities for China's wind power development.

Substrate depletion and microbial community thermal adaptation are major mechanisms that regulate the temperature sensitivity (Q10) of soil microbial respiration. Traditionally, the Q10 of soil microbial respiration is measured using laboratory incubation, which has limits in the continuous input of available substrates and the time scale for microbial community thermal adaptation. How the available substrate and the soil microbial community regulate the Q10 of soil microbial respiration under natural warming conditions remains unclear. To fill this gap in knowledge, a long-term field experiment was conducted consisting of two years of soil respiration observations combined with a soil available substrate and microbial community thermal adaptation analysis under seasonal warming conditions. The Q10 of soil respiration was calculated using the square root method, and it was more affected by the available substrate than by microbial community thermal adaptation. Fertilization management has a stronger effect on soil available substrate than temperature. As the temperature increased, NH4-N proved itself to be important for the bacterial community in the process of Q10 regulation, while dissolved organic carbon and nitrogen were key factors for the fungal community. Based on the niche breadth of microbial community composition, the changing Q10 of the soil respiration was not only closely associated with the specialist community, but also the generalist and neutralist communities. Furthermore, bacterial community thermal adaptation primarily occurred through shifts in the abundances of specialists and neutralists, while changes in species richness and species replacement occurred for the fungal generalists and neutralists. This work indicates that changing available nitrogen and DOC primarily caused by fertilization management contributed more in regulating the Q10 of soil microbial respiration than microbial community thermal adaptation, and there are different mechanisms for bacterial and fungal community thermal adaptation under warming.

This study aimed to investigate the safety and feasibility of a novel stent-assisted in situ intestinal bypass for low-mid rectal cancer patients. Patients who were diagnosed with rectal cancer and received laparoscopic low anterior rectal resection plus a novel stent-assisted in situ intestinal bypass were respectively included from March 2022 to June 2022. Biofragmentable intestinal stent with a protective sleeve was placed in the proximal colon before anastomosis, and feces could be discharged through the protective sleeve without touching the anastomosis, which achieved an in situ bypass of feces. Perioperative characteristics and short-term outcomes were collected. Rectal imaging was performed each week after surgery for the first 3 weeks to surveil the stent and feces delivery. Follow-ups were conducted for more than 3 months. Thirty patients who successfully received surgery were included in this study. There were 18 (60.0%) males and 12 (40.0%) females. As for perioperative characteristics, operation time was 213.8 ± 43.0 minutes, blood loss was 53.3 ± 24.6 mL, time to first flatus via protective sleeve after surgery was 3.2 ± 1.1 days, postoperative hospital stay was 11.8 ± 1.6 days, and time to discharge stent was 22.4 ± 3.2 days. As for short-term outcomes, 6 patients suffered from pneumonia, urinary tract infection or incision infection. During the follow-up, there was no anastomotic leakage or mortality. This novel stent-assisted in situ intestinal bypass is safe and feasible, it might be an applicable way to prevent postoperative anastomotic leakage for patients with low-mid rectal cancer.

Carnobacterium maltaromaticum was found to be specifically depleted in female patients with colorectal cancer (CRC). Administration of C. maltaromaticum reduces intestinal tumor formation in two murine CRC models in a female-specific manner. Estrogen increases the attachment and colonization of C. maltaromaticum via increasing the colonic expression of SLC3A2 that binds to DD-CPase of this bacterium. Metabolomic and transcriptomic profiling unveils the increased gut abundance of vitamin D-related metabolites and the mucosal activation of vitamin D receptor (VDR) signaling in C. maltaromaticum-gavaged mice in a gut microbiome- and VDR-dependent manner. In vitro fermentation system confirms the metabolic cross-feeding of C. maltaromaticum with Faecalibacterium prausnitzii to convert C. maltaromaticum-produced 7-dehydrocholesterol into vitamin D for activating the host VDR signaling. Overall, C. maltaromaticum colonizes the gut in an estrogen-dependent manner and acts along with other microbes to augment the intestinal vitamin D production to activate the host VDR for suppressing CRC.

Transmission line inspection plays an important role in maintaining power security. In the object detection of the transmission line, the large-scale gap of the fittings is still a main and negative factor in affecting the detection accuracy. In this study, an optimized method is proposed based on the contextual information enhancement (CIE) and joint heterogeneous representation (JHR). In the high-resolution feature extraction layer of the Swin transformer, the convolution is added in the part of the self-attention calculation, which can enhance the contextual information features and improve the feature extraction ability for small objects. Moreover, in the detection head, the joint heterogeneous representations of different detection methods are combined to enhance the features of classification and localization tasks, which can improve the detection accuracy of small objects. The experimental results show that this optimized method has a good detection performance on the small-sized and obscured objects in the transmission line. The total mAP (mean average precision) of the detected objects by this optimized method is increased by 5.8%, and in particular, the AP of the normal pin is increased by 18.6%. The improvement of the accuracy of the transmission line object detection method lays a foundation for further real-time inspection.

Pedestrian and vehicle detection plays a key role in the safe driving of autonomous vehicles. Although transformer-based object detection algorithms have made great progress, the accuracy of detection in rainy scenarios is still challenging. Based on the Swin Transformer, this paper proposes an end-to-end pedestrian and vehicle detection algorithm (PVformer) with deraining module, which improves the image quality and detection accuracy in rainy scenes. Based on Transformer blocks, a four-branch feature mapping model was introduced to achieve deraining from a single image, thereby mitigating the influence of rain streak occlusion on the detector performance. According to the trouble of small object detection only by visual transformer, we designed a local enhancement perception block based on CNN and Transformer. In addition, the deraining module and the detection module were combined to train the PVformer model through transfer learning. The experimental results show that the algorithm performed well on rainy days and significantly improved the accuracy of pedestrian and vehicle detection.

Objectives: Although a recent study reported that stimulator of interferon genes (STING) in macrophages has an important regulatory effect on liver ischemia-reperfusion injury (IRI), the underlying mechanism of STING-dependent innate immune activation in liver macrophages (Kupffer cells, KCs) remains unclear. Here, we investigated the effect of STING on liver macrophage pyroptosis and the associated regulatory mechanism of liver IRI. Methods: Clodronate liposomes were used to block liver macrophages. AAV-STING-RNAi-F4/80-EGFP, an adenoassociated virus (AAV), was transfected into the portal vein of mice in vivo, and the liver IRI model was established 14 days later. In vitro, liver macrophages were treated with STING-specific siRNA, and a hypoxia-reoxygenation (H/R) model was established. The level of STING was detected via Western blotting (WB), RT-PCR, and immunostaining. Liver tissue and blood samples were collected. Pathological changes in liver tissue were detected by hematoxylin and eosin (H&E) staining. Macrophage pyroptosis was detected by WB, confocal laser scanning microscopy (CLSM), transmission electron microscopy (TEM), and enzyme-linked immunosorbent assay (ELISA). The calcium concentration was measured by immunofluorescence and analyzed with a fluorescence microplate reader. Results: The expression of STING increased with liver IRI but decreased significantly after the clodronate liposome blockade of liver macrophages. After knockdown of STING, the activation of caspase 1-GSDMD in macrophages and liver IRI was alleviated. More interestingly, hypoxia/reoxygenation (H/R) increased the calcium concentration in liver macrophages, but the calcium concentration was decreased after STING knockdown. Furthermore, after the inhibition of calcium in H/R-induced liver macrophages by BAPTA-AM, pyroptosis was significantly reduced, but the expression of STING was not significantlydecreased. Conclusions: Knockdown of STING reduces calcium-dependent macrophage caspase 1-GSDMD-mediated liver IRI, representing a potential therapeutic approach in the clinic.

The global land area devoted to rubber plantations has now reached 13 million hectares, and the further expansion of these rubber plantations at the expense of tropical forests will have significant adverse effects on the ecological environment. Rubber-based agroforestry systems are considered a preferable approach for ameliorating the ecological environment. Many researchers have focused on the positive effects of rubber-based agroforestry systems on the ecological environment, while ignoring the risks involved in the establishment of rubber-based agroforestry systems. The present study investigated the effects of different-aged rubber-based agroforestry systems on the abundance and diversity of ground arthropods. It has been observed that the abundance and taxon richness of ground arthropods generally showed no difference when comparing young and mature rubber plantations. The rubber-based agroforestry systems significantly decreased the understory vegetation species, along with the abundance and taxon richness of ground arthropods compared to the same aged-rubber monoculture plantations. In addition, the change in the abundance and taxon richness of ground arthropods was greatly affected by the understory vegetation species and soil temperature. The abundance and taxon richness of ground arthropods decreased with the decrease in number of species of understory vegetation. The study results indicate that the establishment of rubber-based agroforestry systems have adversely affected the abundance and richness of ground arthropods to an extant greater than expected. Therefore, single, large rubber-based agroforestry systems are not recommended, and the intercropping of rubber and rubber-based agroforestry systems must be designed to promote the migration of ground arthropods between different systems.

BACKGROUND: Laparoscopic splenectomy (LS) being used after Transjugular intrahepatic portosystemic shunt (TIPS) has not been reported. This report aims to explore the feasibility, safety, and potential efficacy of LS after TIPS hypersplenism secondary to portal hypertension (PHT). METHODS: We retrospectively reviewed a series of six patients who underwent LS after TIPS for hypersplenism secondary to PHT between 2014 and 2020. The perioperative data and patients' clinical outcomes were recorded. RESULTS: LS was successfully performed in all patients. Hypersplenism was corrected after LS in all six patients. Postoperative prothrombin time, prothrombin activity, international normalized ratio, and total bilirubin showed a trend toward improvement. The preoperative and 1-month postoperative albumin and activated partial thromboplastin levels showed no significant difference. Plasma ammonia level and thromboelastography indicators were ameliorated in two limited recorded patients. No postoperative complications such as subphrenic abscess, portal vein thrombosis, variceal bleeding, hepatic encephalopathy, and liver failure occurred during the 1-month follow-up period. CONCLUSION: LS following TIPS is feasible, safe, and beneficial for patients with hypersplenism secondary to PHT. The following LS not only corrects the hypersplenism, but also has the potential to improve liver function.

The accuracy of dryland crop classification using satellite-based synthetic aperture radar (SAR) data is often unsatisfactory owing to the similar dielectric properties that exist between the crops and their surroundings. The main objective of this study was to improve the accuracy of dryland crop (maize and cotton) classification by combining multitype features and multitemporal polarimetric SAR (PolSAR) images in Hebei plain, China. Three quad-polarimetric RADARSAT-2 scenes were acquired between July and September 2018, from which 117 features were extracted using the Cloude-Pottier, Freeman-Durden, Yamaguchi, and multiple-component polarization decomposition methods, together with two polarization matrices (i.e., the coherency matrix and the covariance matrix). Random forest (RF) and support vector machine (SVM) algorithms were used for classification of dryland crops and other land-cover types in this study. The accuracy of dryland crop classification using various single features and their combinations was compared for different imagery acquisition dates, and the performance of the two algorithms was evaluated quantitatively. The importance of all investigated features was assessed using the RF algorithm to optimize the features used and the imagery acquisition date for dryland crop classification. Results showed that the accuracy of dryland crop classification increases with evolution of the phenological period. In comparison with SVM, the RF algorithm showed better performance for dryland crop classification when using full polarimetric RADARSAT-2 data. Dryland crop classification accuracy was not improved substantially when using only backscattering intensity features or polarization decomposition parameters extracted from a single-date image. Satisfactory classification accuracy was achieved using 11 optimized features (derived from the Cloude-Pottier decomposition and the coherency matrix) from 2 RADARSAT-2 images (acquisition dates corresponding to the middle and late stages of dryland crop growth). This study provides an important reference for timely and accurate classification of dryland crop in Hebei plain, China.

The conversion of monoculture rubber (Hevea brasiliensis) plantations into rubber-based agroforestry systems has become a common trend in forestry management in the past few decades. Rubber-Flemingia macrophylla (a leguminous shrub) systems are popular in southwestern China's Xishuangbanna region. The biogeochemical cycles of soil carbon and nitrogen in forests are mainly affected by their fractions. This study investigated the effect of introducing Flemingia macrophylla to rubber plantations of different ages on soil carbon and nitrogen fractions. The experimental treatments included R1 (young rubber plantation), RF1 (young rubber-Flemingia macrophylla system), R2 (mature rubber plantation) and RF2 (mature rubber-Flemingia macrophylla system). The results showed that the introduction of Flemingia macrophylla to rubber plantations of different ages significantly changed soil carbon and nitrogen fractions, improved soil labile organic carbon and nitrogen contents, and ameliorated soil environments. The average soil microbial biomass organic carbon, nitrogen and nitrate-nitrogen in the 0-10 cm soil layer during the experimental period was 38.9%, 55.5%, and 214.7% higher in RF1 than R1, respectively, and 22.1%, 22.2%, and 652.2% higher in RF2 than R2, respectively. Therefore, Flemingia macrophylla can be used as an alternative interplanted tree species within rubber plantations in similar environments of southeastern Asia.

Metformin has been demonstrated to prevent hepatocellular carcinoma (HCC). Metformin acts mainly by phosphorylation of AMPK. However, the phosphorylation status of AMPK and its role in the prediction and prevention of HCC in cirrhotic patients remains unclear. The phosphorylation status of AMPK (Thr172) was determined by immunostaining in tissue microarrays of 426 cirrhotic liver tissues. Low expression of p-AMPK was observed in 94 (22.1%) cases. The median follow-up time was 87 months. HCC occurrence probability at 1/3/5/10 years after Hassab procedure was 3.1/9.6/13.8/30.6% in patients with p-AMPK low expression and 0/0.3/0.3/8% in patients with p-AMPK high expression, respectively. HCC occurrence risk was significantly higher in patients with p-AMPK low expression in univariable analysis (HR, 6.25; 95% CI: 3.36-11.60; P < 0.001) and multivariable analysis (HR, 6.0; 95% CI: 3.24-11.10; P < 0.001). An independent external cohort validated the significance of p-AMPK low expression. In addition, in vivo experiments demonstrated that AMPK activation status was negatively related to HCC occurrence and blocking autophagy by chloroquine counteracted the protective effect of AMPK phosphorylation. These results present novel insight into a critical predictive role of AMPK activation in hepatocarcinogenesis and AMPK activation seems to be a potential target for the prevention of hepatocellular carcinoma in patients with liver cirrhosis.

Hematite is an earth-abundant and ubiquitous semiconductor with a suitable bandgap of 2.1 eV for solar water splitting. Unfortunately, it suffers from a low conduction band position compared to the H+/H2 potential and typically an external bias has to be applied. Here, we demonstrate carbon dot-hematite (CD-Fe2O3) nanocomposites as photocatalysts for visible-light-driven overall water splitting without any external bias or scavenger. Notably, the CD-Fe2O3 nanocomposites (carbon dots, 5 wt%) show a hydrogen evolution rate of 0.390 µmol h-1 and an oxygen evolution rate of 0.225 µmol h-1 under visible light illumination. In our system, carbon dots have been well coupled with hematite and are detected to generate a photo-induced potential. This photo-potential can be combined with hematite to meet the requirement for overall water splitting. In addition, carbon dots can significantly improve the charge separation efficiency. Our finding may greatly enhance the practical application of hematite for solar water splitting.

Chirality has attracted extensive attention in many fields ranging from chemistry to life sciences. Carbon dots (CDs) with good biocompatibility and unique photochemical properties have become a new star in the nanocarbon family. Endowed with chirality, CDs will exhibit more marvellous properties and bridge the fields of material chemistry and life sciences tightly. Herein, we report a facile one-step alkali-assisted electrochemical method to fabricate chiral CDs from cysteine (cys). We showed the chiral evolution of CDs with highly symmetrical circular dichroism (CD) signals in the range from 205 to 350 nm. These chiral CDs have been further demonstrated to be capable of tuning the activity of laccase: the l-CDs can improve the activity of the enzyme up to 20.2%, whereas the d-CDs decrease the activity to 10.4%. A series of experiments confirm that it is the synergistic effect of nanosize and chirality of CDs that induces the change in the structure of laccase and thus leads to the tuning of the laccase activity.

Multifunctional carbon dots (CDs) with lifetime thermal sensing, nucleolus imaging, and antialgal activity properties were synthesized directly from ascorbic acid aqueous solution by a one-step electrochemical method at room temperature. The as-prepared CDs are responsive to temperature and exhibit an accurate linear response of fluorescence intensity vs. temperature (20-100 °C). These CDs can enter a cell and nucleolus, adsorb on the nucleic acids (DNA and RNA) and the fluorescence intensity of CDs is increased by the adsorption of nucleic acids. In addition, the CDs can inhibit the activity of RuBisCO in Anabaena sp., leading to reducing the growth of Anabaena sp. All these properties make the CDs serve as effective fluorescence-based nanothermometers, nucleolus probes, and antialgal agents.

The clinical effects and safety of release and decompression techniques on nerve roots through percutaneous transforaminal endoscopic discectomy (PTED) while treating patients with central lumbar disc herniation (CLDH) were explored. Patient cases of lumbar and leg pain treated in Bethune International Peace Hospital from July 2013 to October 2015 were collected retrospectively. The patients in these cases received imaging examinations such as computed tomography and magnetic resonance imaging. Among these cases, 37 CLDH patients with no other complications were selected for this study. A total of 22 males and 15 females aged 28-54 years, with an average age of 36.8±1.5 years, were the subjects of the study. Their disease course was from 1 month to 3 years, with a median course time of 1.5 years. Visual analogue scale (VAS), Japanese Orthopaedic Association (JOA) scoring and the MacNab method were used to evaluate treatment effects. After permission from patients or their family members, release and decompression techniques of nerve roots were performed through PTED. All patients had successful surgery. Their average surgery time was 41.3 (25.5-57.1) min. A physician followed-up each patient from 0 to 18 months after surgery, with the average follow-up period of 12.1 months. VAS scoring of lower limbs was 7.95±0.82 before surgery and 2.28±0.35, 3 months after surgery. VAS scoring of lower limbs was 2.06±0.58, 1 year after surgery and 2.12±0.23 at the last follow-up appointment. JOA scoring was 12.6±0.72 before surgery and 20.4±1.08, 3 months after surgery. JOA scoring was 21.1±0.82 1 year after surgery and 21.2±0.36 at the last follow-up. Differences are of statistical significance (P<0.05). There were no complications for any of the cases. One patient did not improve after surgery, so a laminectomy and bone grafting internal fixation were performed. Two patients relapsed after surgery and received laminectomy and bone grafting internal fixation. The total percentage of excellent and good rates was 83.5%. In conclusion, release and decompression techniques on nerve roots using PTED while treating CLDH resulted in a safe, effective and less traumatic outcome with fewer complications and quicker pain relief than alternative treatments. Due to the results of this study, the use of these techniques in treating CLDH should be more widely considered.

Wetlands play an important role in regulating the atmospheric carbon dioxide (CO2 ) concentrations and thus affecting the climate. However, there is still lack of quantitative evaluation of such a role across different wetland types, especially at the global scale. Here, we conducted a meta-analysis to compare ecosystem CO2 fluxes among various types of wetlands using a global database compiled from the literature. This database consists of 143 site-years of eddy covariance data from 22 inland wetland and 21 coastal wetland sites across the globe. Coastal wetlands had higher annual gross primary productivity (GPP), ecosystem respiration (Re ), and net ecosystem productivity (NEP) than inland wetlands. On a per unit area basis, coastal wetlands provided large CO2 sinks, while inland wetlands provided small CO2 sinks or were nearly CO2 neutral. The annual CO2 sink strength was 93.15 and 208.37 g C m-2 for inland and coastal wetlands, respectively. Annual CO2 fluxes were mainly regulated by mean annual temperature (MAT) and mean annual precipitation (MAP). For coastal and inland wetlands combined, MAT and MAP explained 71%, 54%, and 57% of the variations in GPP, Re , and NEP, respectively. The CO2 fluxes of wetlands were also related to leaf area index (LAI). The CO2 fluxes also varied with water table depth (WTD), although the effects of WTD were not statistically significant. NEP was jointly determined by GPP and Re for both inland and coastal wetlands. However, the NEP/Re and NEP/GPP ratios exhibited little variability for inland wetlands and decreased for coastal wetlands with increasing latitude. The contrasting of CO2 fluxes between inland and coastal wetlands globally can improve our understanding of the roles of wetlands in the global C cycle. Our results also have implications for informing wetland management and climate change policymaking, for example, the efforts being made by international organizations and enterprises to restore coastal wetlands for enhancing blue carbon sinks.

The nucleotide-binding and oligomerization domain-like receptor 3 (NLRP3) inflammasome participates in the pathogenesis of acute liver injury during sepsis. Bone marrow mesenchymal stem cells (BMSCs) attenuate sepsis through prostaglandin E 2 (PGE2) by increasing the interleukin-10 (IL-10) production of macrophages; moreover, NLRP3 inflammasome assembly is effectively regulated by IL-10 during infection. Whether BMSCs have an effect on the activation of the NLRP3 inflammasome and its underlying mechanism is unclear. Administering of BMSCs to mice or KCs after LPS stimulating have improved liver function and reduced activation of NLRP3 inflammasome in KCs. The beneficial effect of BMSCs was enhanced by over-expression of PGE2 and eliminated by silence of PGE2. Additionally, The IL-10 levels in the serum and supernatant were increased by given BMSCs and further increase by PGE2 over-expressed BMSCs, but decreased markedly by PGE2 silenced BMSCs. Furthermore, extracellular signal-regulated kinase 1 (ERK1) inhibitor reduced IL-10 production in KCs and blocked the inhibitory effect of PGE2 on the activation of the NLRP3 inflammasome. Our data reveal a novel mechanism of BMSC-mediated suppression of the activation of KCs through the secretion of PGE2 by BMSCs, which promotes KCs to secrete IL-10, leading to the inhibition of the NLRP3 inflammasome in KCs.

FcγRIIIa (CD16) is a low-affinity Fc receptor of IgG. As the idio-binding receptor of IgG Fc, it plays an important role in the antibody-dependent cellular cytotoxicity of natural killer cells. The aim of the present study was to investigate the distribution of Kupffer cells (KCs) and the expression of their surface receptor FcγRIIIa in hepatocellular carcinoma. Furthermore, we also aimed to observe the functional mechanism of FcγRIIIa. Immunohistochemical analysis was employed to study KCs and FcγRIIIa. In order to explore the role of FcγRIIIa in the growth of cancer cells, KCs and H22 tumor cells were co-cultured in different serum. The mRNA expression levels of tumor necrosis factor (TNF)-α and FcγRIIIa were analyzed by RT-qPCR; the TNF-α and FcγRIIIa protein expression levels were examined by enzyme­linked immunosorbent assay and western blot analysis, respectively. Our results showed that the number of Kuppfer cells in cancerous tissues (21.6±7.8) was lower than those in para-cancerous (68.8±9.1) tissues and adjacent normal hepatic tissues (62.0±1.9) (P<0.01); this decreased with the reduction in the differentiation degree of cancer (P<0.05). FcγRIIIa-positive cells were similar in morphology to KCs, and their distributive tendency was coincident (P<0.05). The increase in CD16a mRNA levels in the group treated with immune serum was 3.9-, 4.9- and 3.9-fold greater than that in the ordinary serum group at different time points, and CD16a protein expression also markedly increased (P<0.05). However, these effects were inhibited by the addition of anti-IgG Fc serum (P<0.05). The results of the present study suggested that FcγRIIIa resided in KCs, and it contributed to the inhibition of the growth of liver tumor cells.

Liver X receptors (LXRs) in the nucleus play important roles in lipid metabolism and inflammation. The mechanism of LXR regulation of the LPS-induced Toll-like receptor 4 (TLR4) inflammatory signaling pathway remains to be elucidated. C57/BL6 mice were randomly divided into four groups: control, T0901317 (a LXRs agonist), LPS and T0901317+LPS. Additionally, Kupffer cells isolated from male C57/BL6 mice were divided into the same four groups. A decreased amount of inflammatory cells infiltrated the portal areas and the hepatic sinusoids in the livers of mice in the T0901317+LPS group than in those of mice in the LPS group. In the T0901317+LPS group, the serum levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), and tumor necrosis factor alpha (TNF-α) were lower, while the serum level of interleukin-10 (IL-10) was higher. In vitro, Kupffer cells pretreated with T0901317 for 24h presented reduced TNF-α, interferon-beta (IFN-ß) and interleukin-1 beta (IL-1ß) levels, while the IL-10 level increased; however, the mRNA and protein expression levels of interferon regulatory factor 3 (IRF3) and glucocorticoid receptor-interacting protein 1 (GRIP1) were not significantly reduced. The co-IP data illustrated that LXRα bound to GRIP1 specifically in the T0901317+LPS group, while less IRF3 was bound to GRIP1 in the T0901317+LPS group than in the LPS group. Furthermore, the DNA-binding activity of NF-κB was decreased by pretreating Kupffer cells with T0901317 for 24h. These results suggest that activated LXRα competes with IRF3 for GRIP1 binding, thus repressing IRF3 and NF-κB transcriptional activity and inhibiting the inflammatory response initiated by LPS in Kupffer cells.
